Research theme 07

Tumour Evolution, Clonality & Resistance

Subclonal structure, tumour evolution, resistance mechanisms, treatment exposure and dynamic molecular changes.

A researcher at a microscope in a laboratory.

Proposed research direction A direction KPCIRC proposes to explore. Not yet an established programme.

Cancers change, especially under treatment. Resistance can emerge from new alterations or from the expansion of existing subclones. This theme proposes research into how sequential reports, treatment history and clonal structure can be read together.

Questions of interest

  • Which patterns in sequential reports suggest emerging resistance?
  • How should treatment exposure be recorded so molecular change can be interpreted?
  • What can and cannot be inferred about clonal structure from routine reports?

Approaches

  • Timeline-based review of sequential molecular reports
  • Literature curation of resistance mechanisms
  • Structured capture of treatment history

Connection to clinical services

Connects to Progression Review and to resistance and bypass considerations in the SynerGX layer.

Limitations

  • Clonal inferences from routine panels are approximate.
  • A resistance mechanism described in the literature may not explain an individual case.
