Research theme 04
Transcriptomics & RNA Expression
RNA sequencing, expression analysis, pathway activity and molecular signatures.

Proposed research direction A direction KPCIRC proposes to explore. Not yet an established programme.
DNA shows what has changed in the genome; RNA shows which genes are being expressed. Expression data can reveal pathway activity, gene fusions and molecular signatures that DNA alone may miss. This theme explores how expression evidence could complement genomic interpretation.
Questions of interest
- When does RNA-level evidence change the interpretation of a DNA finding?
- How portable are expression signatures across platforms and tumour types?
- How should fusion and expression results be presented alongside DNA variants?
Approaches
- Pathway-level interpretation of expression results
- Cross-platform comparison of signatures from published data
- Integration of fusion findings into structured evidence reviews
Connection to clinical services
Supports the biological-context reading in the PhenoMap layer when expression data are part of a supplied report.

Limitations
- Expression signatures validated in one setting may not transfer to another.
- A pathway signal does not by itself establish treatment benefit.
